Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Agriculture - Department of Pomology Summary: This study was conducted to investigate the ability to produce whole plantlets from immature female inflorescences of date palm trees (Amhat cv.)through tissueCulturestechniq. The best significant result of sterilization that achieved the highest alivespathes was obtained on 30th January and double sterilization with clorex at 50% then 40% each for 30 minute. MS medium supplemented with 10 mg 2,4-D + 3mg 2 ip + 1 mg PBZ resulted in the highest flowery bud elongation . Meanwhile the same medium MS contained PBZ at 2 mg gave a positive result for percentage swelling ,browning and callus, As for production direct somatic embryo, MS medium with added TDZ at 1 mg/L + 0.5 mg/L NAA gave the best result beside the lowest browning percentage. The nitrogen sources NH4 NO3 and KNO3 at 0.83 : 1.43 g/L results in the lowest browning and vitrification degree of friable callus explants. Moreover, this ratio produced the highest globularization degree as well as the highest value of germination percentage and growth rate. Also adding glutamine at 400 mg/L enhanced germination and growth rate of friable callus. MS medium at {u00BD} strength and supplemented with 40 mg/L sucrose produced the highest germination percentage of somatic embryo. MS medium with added 5g/L PEG produced plantlets with high length , root number and root length. After second subculture PEG at 8 g/L produced the highest number of secondary roots as well as it gave the highest percentage survival at acclimatization stage
