Background & objectives: The identification of human body fluids or tissues through mRNA-based profiling is very useful for forensic investigations. Saliva used for this study as it is a mirror of the body health and easy sampled by non invasive methods. Aim of this study: Definitive identification of human saliva at different time intervals, using mRNA markers histatin3 (HTN3) & statherin (STATH), by reverse transcriptase PCR (RT PCR).Results: The mean value of the age of the candidates was 29 years old ± 4.06. Males were 55% and females were 45% of all studied groups. There was no statistical significant difference regarding the gender or age variables. Both HTN3 & STATH were highly specific to fresh saliva samples while on the other hand they were not found in fresh blood samples. Mean values of HTN3 and STATH were higher at 1 month and lower at six months in both cotton group and rolling paper groups. There was high significance difference between the HTN3 & STATH in cotton group than in rolling paper group. There was a highly significant strong-negative correlation between levels of genes (HTN3 & STATH) and different time intervals (one, three and six months) in both groups. There was a significance association between males and females regarding the HTN3 values at one month & at three months only
