This study was carried out to assess the feasibility of generating insulin-producing cells from progenitor cells obtained from the bone marrow (BM) and cord blood (CB) and compare the trans-differentiation potential of the two studied sources (BM versus CB cells). I- In this study, 20 samples of umbilical cord blood were obtained, isolation of mesenchymal stem cells (MSCs) was done. In vitro expansion of MSCs was performed. MSCs were propagated in-vitro, the insulin-producing cells differentiation medium was added. The cells showed a change in their morphology insulin-producing cells appeared. II- Twenty samples of rat bone marrow were obtained; isolation of mesenchymal stem cells (MSCs) was done. In vitro expansion of MSCs was performed. MSCs were propagated in vitro, the insulin-producing cells differentiation medium was added. The cells showed a change in their morphology, and became insulin-producing cells appeared. III. RNA extraction was performed; RNA was transformed to (cDNA) and expression of GAPDH, Insulin genes were assessed using RT-PCR technique
