With the emergence of multidrug-resistant bacteria, the search for new antimicrobial drugs has become of utmost importance. In the present study, 16 soil samples were collected from different regions in Egypt. Forty-nine strains of actinomycetes were isolated and tested for antibacterial activity.The isolates were screened primarily against Staphylococcus aureus and Escherichia coli. Four isolates inhibited one of the tested microorganisms while two isolates inhibited both test organisms. Only one isolate (A5) that showed potent antibacterial activity against both tested microorganisms was chosen for further characterization and identification.16S rRNA gene sequence of the isolate (A5) was submitted to genbank with deposition number MF124800.Using BLAST the isolate (A5) was identified as Streptomyces aureofaciens.Starch nitrate broth for 5 days at pH 7 and 30 C temperature were the best parameters that induce maximum production of antibacterial metabolite with broad spectrum. Mass Spectrum showed that the molecular weight of the isolated metabolite is above 700
