Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Pharmaceutics
The objective of this thesis is to develop and formulate a new potential radioiodine and technetium-99m labeled chemotherapeutic agents for tumor imaging. The choice of the studied anticancer agent MPD depended on suitability of its mechanism for tumor targeting. Moreover, it also depended on the suitability of its chemical structure. Part one describes the procedures of MPD microemulsion (MPDME) systems preparation using water titration technique and MPS solution (MPDS). Characterization and optimization of MPDME formulae were performed. A 3122 full factorial design using Design-Expert® software was adopted to investigate the effect of different system parameters on ME characteristics and to identify the optimum system. Part two describes the proceduresto evaluate A10, MPD and 125I-MPD as HDAC8 inhibitors by molecular docking. It describes the procedures of radiolabeling of MPDME1, MPDS and placebo with 125I via direct electrophilic substitution reaction using Chloramine-T (CAT) as an oxidizing agent. Part three describes the evaluation of binding affinity of MPD as well as 99mTc-MPD with the DNA using molecular docking
