Thesis (Ph.D.) - Cairo University - Faculty of Science - Department of Botany - Microbiology
This study was undertaken to achieve the following objectives: The present work aims to fulfill six major goals. Potential application of yeast isolates for extracellular BS production by using different ten screening methods in the primary screening. Then molecular identification of significant yeasts involved in efficient BS production by 18S rRNA gene amplification and sequencing analysis tools. Thereafter,Optimization of BS production by the most potent yeast strain, regarding the effect of carbon and nitrogen sources, C/N ratios, pH values, temperature in shake-flask method and BS production scaling up in batch bioreactor. Further more, the resulting crude BSs isolation and purification including solvent extraction and preparative HPLC, characterization and identification by TLC, GC-MS, FTIR and LC-MS/MS as primary essentials to search and reveal their chemical nature and possible structures. Finally, the influence of these biocompounds on the phytopathogenic fungi as antimicrobial agents was in vitro evaluated
