| Summary, etc. | In this study, precise gene editing techniques was applied to generate wheat 3'(2'), 5'-bisphosphate nucleotidase (Sal1) mutants using a multiplex sgRNA-CRISPR/Cas9 genome editing system. Five active Sal1 homologous genes were found in the genome of Giza168 in addition to another apparently inactive gene on chromosome 4A. Three gRNAs were designed and used to target exons 4, 5 and 7 of the five wheat Sal1 genes. Among the 120 Giza168 transgenic plants, 41 lines exhibited mutations and produced heritable sal1 mutations in the M1 progeny, and 5 lines were full 5 gene knock-outs. These mutant plants exhibit a rolled-leaf phenotype in young leaves and bent stems, but there were no significant changes in the internode length and width, leaf morphology and stem shape. Anatomical and scanning electron microscope studies of the young leaves of mutated sal1 lines showed closed stomata, increased stomata width and an increase in the size of the bulliform cells with rolled. The sal1 mutant seedlings germinated and grew better on media containing polyethylene glycol than wildtype seedlings. The obtained results indicate that the application of the multiplex sgRNA-CRISPR/Cas9 genome editing is an efficient tool for mutating more multiple Sal1 loci in hexaploid wheat. |

| Summary, etc. | في هذه الدراسة، تم استخدام تقنيات التعديل الجينى لاحداث طفرات لجين3'(2'),5'- Bisphosphate Nucleotidase (Sal1) في القمح و ذلك باستخدام نظام لتعديل الجينى المتعدد sgRNA-CRISPR / Cas9 . وقد أوضحت النتائج وجود خمسة جينات متماثلة تعبر عن نفسها من نوع Sal1 في جينوم صنف القمح جيزة 168 بالإضافة إلى جين آخر صامت على الكروموسوم 4A. تم تصميم ثلاثة gRNAs وإستخدامها لإستهداف ثلاث مناطق مشفرة هما 4 و5 و 7 من الخمسة جينات الخاصة ب Sal1 للقمح. وقد أوضحت النتائج ظهور 41 طفرة مختلفة من بين 120 نبات معدلاً وراثيًا في جيزة 168، وأنتجت طفرات Sal1 قابلة للوراثة في النسل M1 و 5 سلالات تحتوي على 5 جينات حدث لها فقد كامل للوظيفة. تُظهر هذه النباتات الطافرة نمطًا ظاهريًا للأوراق الملفوفة في الأوراق الصغيرة والسيقان المنحنية، ولكن لم تكن هناك تغييرات كبيرة في الطول والعرض الداخلي، ومورفولوجيا الأوراق وشكل الساق. أظهرت الدراسات التشريحية والمسح بالميكروسكوب الإلكتروني للأوراق الصغيرة لطفرات Sal1 المتحولة ثغورًا مغلقة، وزيادة في عرض الثغور وزيادة في حجم الخلايا اللفة. شتلات طافرة Sal1 نمت بشكل أفضل على وسط يحتوي على من البولي إيثيلين جلايكول. تشير نتائجنا إلى أن تطبيق التعديل الجينى المتعدد sgRNA-CRISPR / Cas9 هو أداة فعالة لتحور المزيد من مواقع Sal1 المتعددة في القمح سداسي المجموعة الكروموسومية. |
