Summary, etc. | Introduction: the growth of Dunaliella salina SA20 (Dunal) Teodoresco Egyptian <br/>isolate (from El-Bardawil Lake, Sinai, Egypt) in wastewater such as mozzarella cheese <br/>whey (MCW) is a more cost-effective cultivation method and growing D. salina SA20 <br/>in such waste could be used safely in biotechnological applications. <br/>Methodology: the use of 20% v/v MCW/Loeblich nutrient medium as growth <br/>supplement allowed mixotrophic nutrition, while growth in 100% Loeblich allowed for <br/>autotrophic nutrition. Mixotrophy and autotrophy were compared utilizing the next-<br/>generation sequencing technique, transcriptome assembly, and differential gene <br/>expression analysis using RNA-Seq. <br/>Results: the optimum growth performance of D. salina SA20 cells was observed in pH <br/>7.5 and was indicated by measured values of chlorophyll a (10.51 mg/L), dry weight <br/>(405 mg/L), cell count (6.92 x 106 cell/ml), and optical density (1.12). Additionally, a <br/>growth curve experiment comparing the mix- and autotrophic conditions presented <br/>higher values of the relative mean and maximum growth rates in mixotrophic <br/>conditions than in the autotrophic conditions. The pathway enrichment analysis under <br/>mixotrophic conditions found down-expression in important genes for nitrogen <br/>metabolism, carbon metabolism, purine and pyrimidine biosynthesis, and up-expression <br/>in genes encoding for enzymes involved in photosynthesis, carbon fixation, <br/>glucogenesis, and ribosome production pathways. <br/>Conclusion: the results might suggest a connection between the induced growth strategy <br/>used in mixotrophy and improved photosynthesis coupled with enhanced <br/>glucogenesis. |

Summary, etc. | تم استخدام شرش الموتزريلا والناتج بكميات كبيرة من صناعة الألبان كوسط غذائي لنموطحلب دوناليلا سالينا بطريقة آمنة في تطبيقات التكنولوجيا الحيوية وذلك بإضافته إلى الوسطالغذائي الغير عضوي )لوبلش( بتركيزات مختلفة) 20 و 50 و 75% شرش/لوبلش( كمثالللتغذية المختلطة بينما يعتبر النمو ف ي الوسط الغير عضوي )١٠٠% لوبلش( هو مثال للتغذيةالذاتية وقد دلت النتائج على ان التغذية المختلطة بتركيز ٢٠% قد سجلت أعلى النتائج )كلوروفيلأ 10.51 مجم/لتر والوزن الجاف 405 مجم/لتر وعدد الخلايا 6.92× 610 خلية/مل(، والكثافةالبصرية) 1.12(. كما تم استخدام قيم مختلفة للاس الهيدروجين ي لدراسة تأثير تلك القيم المختلفة حيث وجد انpH 7 هو القيمة المثلى للنمو. بالإضافة إلى ذلك، قدمت تجربة منحنى النمو التي تقارن بين ظروف التغذية المختلطة والتغذية الذاتية قيمًا أعلى للمتوسط النسبي ومعدلات النمو القصوى في ظروف التغذية المختلطة) 0.19, 0.21 /يوم، على التوالي( مقارنة بظروف التغذيةالذاتية) 0.16، 0.18/ يوم، على التوال ي(. كشف تحليل الترنسكربتوم عن إفراط في التعبير في الجينات المشاركة في التمثيل الضوئي، وتثبيت الكربون، وتكوين الجلوكوز، ومسارات تخليق الريبوسوم ،وتثبيط في مسارات النيتروجين، وأيض الكربون، والتخليق الحيوي للبيورينوالبيريميدين. |
