Salinity is one of the most damaging abiotic stress factors in agriculture, it has a negative impact on crop growth, production, and development. two genes AtLTP1 and LeCBF1 from Arabidopsis Thaliana and Solanum Lycopersicum were cloned and sequenced. These genes were cloned into the pYES2 vector for Saccharomyces cerevisiae expression. After being exposed to increasing concentrations of NaCl (0, 1.7, 1.8, 1.9, 2.0, 2.1, 2.2, and 2.3M) for 7 days, transgenic yeast was tested for their ability to survive under salt-stress conditions and their growth response. Afterward, the yeast growth was measured at OD600nm in a spectrophotometer. The growth of the transgenic yeast and the control showed clear disparities in the results. The ability of yeast to survive salt stress depended on the salt concentration, which was 1.8M NaCl. The mean growth values for the transgenic yeast carrying the AtLTP1 and LeCBF1genes were 1.6386 c and 1.5100 cd respectively, at this concentration as opposed to 0.8553 fg for the control. Additionally, the growth of the control cells was dramatically hindered when the salt content was increased to 1.9 M NaCl. However, the two transgenic genes in yeast cells continued to grow well, though at a slower rate, up to 2.3 M NaCl. The two genes' expression in transgenic yeast in response to salt stress was verified by RT-PCR. In this transgenic yeast, the precise AtLTP1 and LeCBF1 primers successfully amplified the genes at 368 base pairs and 633 base pairs, respectively.<br/>In addition to the transgenic yeast's growth response to salt stress, the findings<br/>showed that increasing salinization level considerably boosted the transgenic yeast’s intracellular proline accumulation. |

الملوحة من اخطر الضغوط البيئية علي الزراعة وتؤثر سلبا علي نمو المحاصيل وتطورها وانتاجيتها. تم كلونة وتحديد تتابع النيكليوتيدات الخاص بجينين LeCBF1 <br/> و AtLTP1 وقد تم تطعيم هذة الجينات علي ناقل التعبير الجيني pYES و نقلها الي <br/>الخميرة Saccharomyces cerevisiae و تم تقييم قدرة الخمائر المحورة وراثيا <br/>للبقاء حية تحت ظروف الاجهاد الملحي حيث تم تقدير استجابتها للنمو وبزيادة مستوي الملوحة من كلوريد الصوديوم بتركيزات (.و1.7و1.8 و1.9و2.0 2.2 و2.1و و2.3 مولر) لمدة 7 ايام عند طول موجي 600نانوميتر.واظهرت النتائج اختلافات في النمو بين الخمائر المحورة وراثيا والغير محورة وراثيا. وقد اظهرت النتائج ان تركيز 1.8 مولر كلوريد الصوديوم يعتبر التركيز الحرج لتحديد قدرة الخميرة علي تحمل الاجهاد الملحي. حيث وجد ان عند هذا التركيز متوسط نمو الخلايا المحورة وراثيا باي من الجينين<br/> AtLTP1 و LeCBF1 كان 1.636و1.5100 علي التوالي مقارنة ب0.8555 للخلايا المحورة وراثيا بالبلازميد فقط. علاوة علي ذلك وجد ان زيادة تركيز كلوريد الصوديوم الي 1.9 مولر ادي ذلك الي تثبيط لنمو الخلايا المحورة وراثيا بالبلازميد فقط بينما الخلايا المحورة وراثيا باي من الجينين استمرت في النمو حتي تركيز 2.3 مولر كلوريد الصوديوم ولكن مع انخفاض مستوي النمو. وتم تاكيد التعبير الجيني للجينين لاستجابتها للاجهاد الملحي في الخميرة المحورة وراثيا باستخدام تفاعل البلمرة المتسلسل ذو النسخ العكسي و نجحت البادئات المتخصصة لكلا الجينين في تضاعف الجينات عند وزن جزيئي و 368و633 زوج قاعدة علي التوالي واظهرت الخميرة المحورة وراثيا زيادة معنوية كما اوضحت النتائج تراكم البرولين داخل الخلايا مع زيادة مستوي الملوحة. |
