Summary, etc. The present study is carried out to evaluate the molluscicidal properties of CuO NPs on Biomphalaria <br/>alexandrina snails, the intermediate host of S. mansoni, and its larval stages. The study included the <br/>effect of the sub-lethal concentrations of CuO NPs on the biological, biochemical, histological and <br/>genotoxicological parameters. The findings are summarized as follows: <br/>1. The present results confirmed the single crystal structure of the CuO NPs by X-ray diffraction <br/>which confirmed its structure type at 196K and room temperature. No characteristic peaks of <br/>any impurities were detected, suggesting that high-quality CuO NPs were synthesized. The <br/>average crystallite size of CuO NPs was found to be around 40 nm. Also, transmission electron <br/>microscopy (TEM) showed typical image of CuO NPs. This picture exhibited that the majority <br/>of the particles were polygonal in shape with smooth surfaces. The UV–VIS spectrophotometer <br/>showed a sharp absorption band. <br/>2. The present experiment confirmed the molluscicidal properties of CuO NPs against adult B. <br/>alexandrina snails after 24 h of exposure followed by another 24 h for recovery. The half lethal <br/>concentration (LC50) of CuO NPs was 40 mg/l. <br/>3. The present results showed that the survival and growth rates of B. alexandrina juvenile snails <br/>exposed to LC0 (4 mg/l), LC10 (15.6 mg/l) and LC25 (27.18 mg/l) of CuO NPs for 24 h/ week <br/>were significantly decreased compared to control snails. This decrease is concentration <br/>dependent, where, increasing the concentration to LC25 caused a sharp decrease in the survival <br/>and growth rates of treated snails. <br/>4. The survival, reproductive and fecundity rates of adult snails exposed to LC0 (4 mg/l), LC10<br/>(15.6 mg/l) or LC25 (27.18 mg/l) CuO NPs were significantly lower than those of the control <br/>group. <br/>5. The current data indicates that CuO NPs have miracidicidal and cercaricidal activities against <br/>S. mansoni miracidia and cercariae and these activities were dependent on both the <br/>76<br/>concentration and the exposure period. Raising the concentration to LC90 caused severe and <br/>sudden death of treated miracidia after 10 min. Also, the mortality rate of cercariae increased <br/>with increasing the concentration of CuO NPs and the period of exposure, where, 100% of the <br/>cercariae death rate occurred within 30 min after exposure to LC90. <br/>6. Scanning electron microscopy (SEM) examination showed that the normal head foot- region <br/>of B. alexandrina snails displayed smooth tegmental surface with conspicuous microvilli and <br/>tentacles have a flat surface with fine cilia and the mantle has a smooth tegmental surface. <br/>Exposure of B. alexandrina snails to the sub lethal concentrations of CuO NPs (LC10 15.6 mg/l <br/>– LC25 27.18 mg/l) for 24 h followed by 24 h recovery caused morphological alterations. <br/>Where, the tentacles revealed rough folds with erosion at its apex. The tegmental surface of <br/>the mantle became turgidity, rough, blebbing, peeling, and tortuosity. By raising the <br/>concentrations of CuO NPs (LC25 27.18 mg/l), the mantle is ruptured and its tegmental surface <br/>showed erosion, tortuosity and nipples. Tentacles ruptured and became more tortuous, with <br/>rough folds. <br/>7. The present results revealed a significant decrease in TAA, GSH, CAT and SOD contents in <br/>tissue after exposure of B. alexandrina snails to the sub-lethal concentrations of CuO NPs <br/>(LC10 15.6 mg/l – LC25 27.18 mg/l) for 24 h. <br/>8. The present investigation revealed histopathological alterations in both the digestive and the <br/>hermaphrodite glands of B. alexandrina snails after exposure to sub-lethal concentrations of <br/>CuO NPs (LC10 15.6 mg/l – LC25 27.18 mg/l) for 24 h followed by another 24 h for recovery. <br/>9. The present study showed DNA damage with the absence and presence of new bands in the <br/>exposed snails to sub lethal concentrations of CuO NPs compared to the control groups. Also, <br/>decrease in genomic stability confirmed DNA damage caused by CuO NPs.

Summary, etc. داء البلهارسيات هو مرض استوائي رئيسي ، ولكن يتم تجاهله بشكل عام ، يحمله القواقع من جنس البيومفلاريا الكسندرينا ، والتي تنتشر بشكل كبير في مصر. و للحد من انتشار داء البلهارسيا طرق عديده منها السيطرة على هذا الحلزون الذى يعتبر العائل الوسيط للبلهارسيا ،كما اكتسبت تكنولوجيا النانو اهتمامًا كبير فى معالجة بعض الامراض. وبالتالي صممت هذه الدراسه لتقييم نشاط جزيئات أكسيد النحاس النانونية كمبيدات للرخويات مثل قواقع بيومفلاريا الكسندرينا على الجوانب البيولوجية و الجزيئية و أيضا العلامات الحيوية الفسيولوجية والنسيجية والسمية الجينية. أظهرت النتائج أن جزيئات أكسيد النحاس النانوية ، تتميز ، بنية بلورية واحدة بمتوسط حجم بلوري حوالي 40 نانومتر .لوحظ التأثير السام لجسيمات أكسيد النحاس النانوية ضد القواقع بعد التعرض للتركيزات تحت المميته (50LCو 90LC كانت 40 , 64.3 مجم / لتر ، على التوالي) مما أدى الى زياده فى مستوى بيروكسيد الدهون (MDA) و أكسيد النيتريك يصاحبه انخفاض فى محتوى الجلوتاثيون وأنشطة الكاتلاز وديسموتاز الصوديوم فى القواقع العرضه مقارنة بالغير معرضه. أيضا اظهرت النتائج انخفاض في معدلات النمو والتكاثر للقواقع المعرضه للتركيزات شبه مميتة ، وكذلك انخفاض في قابلية بقاء البيض .علاوة على ذلك ، أظهرت جزيئيات أكسيد النحاس النانوية تأثيرًا سامًا على المراحل اليرقيه للشيستوسوما .أكدت الدراسه بالفحص المجهري الإلكتروني( SEM) ) للرأس والقدم و البرنس للقواقع المعرضه للتركيزات شبه المميتة (15.610LC25 27.18 - LC مجم / لتر) من CuO NPs إلى تغيرات مورفولوجية في السطح الخارجى. كما حدثت تغيرات نسيجية مرضية في الغدة الهضمية حيث تمزق خلايا الجهاز الهضمي وتفريغها وكان هناك زيادة ملحوظة في عدد الخلايا الإفرازية وكانت هذه التغيرات تعتمد على التركيز مقارنة بالقواقع الغير معرضه. علاوة على ذلك ، تم الكشف عن تلف الحمض النووي في القواقع المعرضه من خلال تغييرات في RAPD profile . و ختاما أظهرت جزيئات أكسيد النحاس النانوية تأثيرًا كبيرًا ضد العوائل الوسيطة لـ S. mansoni مما يجعلنا نتجه الى استغلال الموارد المحلية كمبيدات للرخويات.
