| Summary, etc. | Background: Non-Hodgkin lymphoma (NHL) encompasses a broad spectrum of diseases, from the most benign to the worst forms of cancer. Currently, NHL is considered one of the fastest growing malignant tumors worldwide. The prevalence of NHL has risen in recent decades due to many factors, including immunosuppression, genetics, and chemical exposure. Hematological malignancies are intimately linked to anomalies in DNA repair systems since these mechanisms are important for repairing DNA damage & maintaining genomic integrity.<br/>Aim: This study aimed at studying association between two important genes in DNA repair; RAD51 3879T>A and XRCC3 562A>G genetic variants & B- NHL hazard among Egyptian population.<br/>Subjects: The current research involved 100 Egyptian patients with B-NHL & 100 age & gender matched healthy control subjects.<br/>Methods: All included patients were subjected to clinical, laboratory & radiological assessment in addition to assessment of their reaction to therapy. Genetic typing of patient and control subjects for RAD51 rs2619679 (g.3879T>A) and XRCC3 rs1799796 (c.562A>G) polymorphism was done by means of real time polymerase chain reaction (PCR) utilizing Taqman SNP genotyping.<br/>Results: Comparison between the distribution of alleles and genotypes revealed that neither the existence of A or T allele of the RAD51 gene nor the A or G allele of the XRCC3 had statistical significant difference among B- NHL cases & healthy controls except for AG genotype in XRCC3 which revealed statistical significant variance among B-NHL patients & healthy controls (P value=0.04, OR=2.752, CI %=1.048-7.228) indicating increased risk in individuals carrying the AG genotype to develop NHL.<br/>Also, Statistical analysis of relation between mutant or wild genotypes of RAD51 and XRCC3 in contrast to different clinicopathological data in B NHL patients revealed no statistical significant variance (P value >0.05) except for XRCC3 with hepatomegaly (P value=0.045).<br/>Conclusion: Although most of our study findings are statistically insignificant, the current study helps to shed light on the possible impact of single nucleotide polymorphisms of RAD51 and XRCC3 in B-NHL. |

| Summary, etc. | تشتمل امراض الغدد الليمفاوية الغير هودجيكينية على العديد من الأمراض الليمفاوية ذات الخصائص الاكلينيكيه والخلويه المتنوعة و تنشا هذه الأورام عاده من الخلايا الليمفاوية من النوع ب وبنسبه أقل من الخلايا الليمفاوية من النوع ت. <br/>امراض الغدد الليمفاوية الغير هودجيكينية تعتبر أحد أسرع الأورام الخبيثة نموًا في العالم نتيجة لعدة عوامل منها ضعف المناعة و العوامل الوراثية والتعرض للعوامل الكيميائية و تعتبرآليات إصلاح الحمض النووي مسؤولة عن تصحيح تلف الحمض النووي والحفاظ على سلامة الجينوم ، ومن الواضح أن اضطراب هذه الآليات يرتبط ارتباطًا وثيقًا بسرطانات الدم.<br/>هدفت هذه الدراسة إلى دراسة الارتباط بين المتغيرات الجينية في جينين مهمين في إصلاح الحمض النووي RAD51 و XRCC3و مخاطر سرطان الغدد الليمفاوية الغير هودجيكينية في السكان المصريين و شملت الدراسة مائة مريض مصري و تم الاستعانه بمائة من المتطوعين الأصحاء كمجموعة ضابطه للبحث.<br/>خضع جميع المرضى المشمولين إلى التقييم السريري والمختبري والإشعاعي وتقييم استجابتهم للعلاج و تم إجراء الفحص الجيني لمجموعة المرضى و المجموعة الضابطه للبحث لـ RAD51 rs2619679 و XRCC3 rs1799796 عن طريق تفاعل البلمرة المتسلسل اللحظي <br/>كشفت المقارنة بين توزيع الأليلات والأنماط الجينية أنه لا يوجد فرق إحصائي في أليل A أو T للجين RAD51 ولا أليل A أو G من XRCC3 بين مجموعة المرضى و المجموعة الضابطه باستثناء النمط الجيني AG في XRCC3 الذي أظهر فروق ذات دلالة إحصائية (قيمة P = 0.04). أيضًا ، لم يظهر التحليل الإحصائي للعلاقة بين الأنماط الجينية الطافرة أو البرية لـ RAD51 و XRCC3 و بين البيانات الإكلينيكية المختلفة في مرضى B NHL أي فرق ذي دلالة إحصائية (قيمة P> 0.05) باستثناء XRCC3 مع تضخم الكبد (قيمة P = 0.045) <br/>على الرغم من أن نتائج دراستنا غير ذات دلالة إحصائية ، إلا أن الدراسة الحالية تساعد في إلقاء الضوء على الدور المحتمل لتعدد أشكال النوكليوتيدات المفردة لـ RAD51 و XRCC3 في امراض الغدد الليمفاوية الغير هودجيكينية. |
