Summary, etc. Ficus species (Moraceae) have been utilized for nutrition and traditional medicine, and plants of this family are rich in bioactive natural products. Characterizing the phytochemistry of Ficus species is still challenging due to the inherent complexity of the plant metabolomes and the incomplete decoding of their chemical space. Therefore, in this study, we used molecular networking to clarify the chemical classes present in the leaves of three Ficus species (F. deltoidea Jack, F. drupacea Thunb, and F. sycomorus L.), and molecular networking was used to analyze their chemotaxonomy. Using computational approaches, 90 metabolites from various classes (phenolic acids, flavonoids, furanocoumarins, fatty acids, and terpenoids) were annotated. Phenolic acids were identified as the main class found in the three species. While furanocoumarins were suggested to be present in F. sycomorus, flavonoids-C-glycosides, flavonoids-O-glycosides, and isoflavonoids were the main chemical components of F. drupacea and F. sycomorus. Vomifoliol-based sesquiterpenes were proposed in F. deltoidea. Using SCOT and ISSR markers, the chemotaxonomic difference was consistent with DNA fingerprinting, where F. deltoidea displayed a distinct chemical fingerprint and genotype.<br/> Among the three species, the 70% alcoholic extract of F. drupacea Thunb leaves demonstrated the most potent anti-acid and anti-Helicobacter pylori activities. The active extract was fractionated by Diaion® HP20 column chromatography into 5 fractions; H2O, 25% MeOH/H2O, 50% MeOH/H2O, 75% MeOH/H2O, and 100% MeOH. Among the tested fractions, the Ficus drupacea water fraction (FDWF) was the most active as an anti-acid, anti-H. pylori and anti-urease. So, the gastroprotective effect of FDWF against ethanol-induced peptic ulcer in rats was evaluated. Twenty-three compounds in FDWF were tentatively identified by UPLC-PDA-ESI-MS/MS; the majority of which were phenolic acids and their derivatives. Using LC-ESI-MS/MS, FDWF was standardized to contain 23.66±2.62 mg/g of quinic acid and 8.86±0.29 mg/g of chlorogenic acid. Moreover, molecular interactions of the compounds with the active site of the urease enzyme were supported by the molecular docking analysis where dihydroxybenzoic acid-O-pentoside/hexoside and dihydroxybenzoic acid-O-dipentoside showed the highest binding affinity and very favorable absorption, distribution, metabolism, and excretion (ADME), and drug-likeness properties.
