| Summary, etc. | Structural complexities of peptide and protein therapeutics have significant effects on their safety and efficacy. Antibody-drug conjugates (ADCs) are considered to be fast-growing innovative biopharmaceuticals. The science used for conjugating potent cytotoxic payload to the targeted monoclonal antibody through a chemical linker has played a great value in the area of oncology treatment. Biosimilars are also considered to be a fast growing category among the biopharmaceuticals. Because of the considerable diversity of biosimilar product manufacturing platforms, structural characterization must be considered during the establishment of a biosimilar product development programme. To demonstrate biosimilarity, a meaningful comparative analytical assessment of the proposed biosimilar and the reference product is required, using cutting-edge analytical assay to assess. The degree of heterogeneity, functional properties, impurity profile and degradation profile indicating stability technique should be studied. In this study, stability assessment protocol was tested for two ADCs (Polatuzumab vedotin and brentuximab vedotin) using complementary techniques in addition to introducing a head-to-head stability indicating protocol for the assessment of physicochemical properties of Adalimumab biosimilars. The proposed protocol included stability indicating chromatographic methods developed and validated in terms of selectivity, linearity, and intra- and inter-day variations in addition to peptide mapping identity test. |

| Summary, etc. | تتكون الرسالة من جزئين رئيسسن بالإضافة الى المراجع<br/>الجزء الأول و هو خاص بدراسة الثبات الخاصة بالأجسام المضادة المقترنة بالدواء<br/>القسم الأول: المقدمة<br/>هذا الجزء يتضمن مقدمة عامة عن التعريف بالمستحضرات الحيوية وطرق انتاجها و ما تواجهه هذه الصناعة الصيدلية من تحديات على مستوى التصنيع والرقابة وضمان الجودة وذلك بسبب التعقيدات التي تتسم بها عمليات التصنيع من خطوات التخمير الى خطوات التنقية ثم الصياغة في الشكل الصيدلي الذي يوفر افضل درجات الإتاحة والفاعلية والثبات الصيدلي. كما يشمل خلفية مختصرة عن طبيعة البروتينات المعالجة ونبذة عامة عن الطرق المستخدمة في تحليل المستحضرات الحيوية وأهم محاور ضمان جودتها. ويشمل هذا االجزء مقدمة عن الاجسام المضادة المقترنة بالدواء و التي تم دخولها الي سوق الادوية حديثا. بالاضافه الي مقدمة عن الادوية المستخدمة في هذه الدراسة. <br/>القسم الثاني: التجارب العملية <br/>يشمل هذا الجزء طرق التحليل المستخدمة في هذه الدراسة ويسلط الضوء على كيفية اختيارها لاستغلالها بالطريقة المثلى.<br/>تم استخدام تقنية كروماتوجرافيا السائل ذو الكفاءة العالية لاستبعاد الحجم بعد التحقق من كفاءتها ومدي قدرتها علي فصل المستحضرات المستخدمة من التجمعات البروتينية. كذلك تم استخدام طريقة كروماتوجرافيا السائل ذو الكفاءة العالية في الحالة المعكوسة والتحقق من كفاءتها لتعيين نسبه الدواء الحر للمستحضرات المستخدمة. تم استخدام تقنية ديناميكية تشتت الضوء المكملة لكروماتوجرافيا استبعاد الحجم ووجد انها طريقة حساسة للكميات الصغيرة من الشوائب ذات الوزن الجزيئي العالي. تم التحقق من صحة الفحص المناعي المرتبط بالإنزیم لتأكید النتائج من تقنیة كروماتوجرافیا السائل ذو الكفاءة العالیة لاستبعاد الحجم في التحقق من سلامة الأجسام المضادة. تم استخدام عوامل الضغط المختلفة وقياس مدى تاثيرها علي المستحضرات المستخدمة تحت الدراسة. |
