Summary, etc. Polycystic Ovary Syndrome (PCOS) is a widespread endocrinal disease with no definite cure to date. Fingers are pointed to cyclic adenosine monophosphate (cAMP) as a possible common culprit in both increased steroidogenesis and reduced autophagy in PCOS pathogenesis. Berberine (BBR) is an isoquinolone derivative of natural origin that was used for centuries as an inducer of ovulation. The current study was performed to shed light on the possible effect of BBR in managing PCOS through targeting ovarian cAMP level. Rats were assigned into 4 groups: Control, PCOS induced by 1mg/kg oral letrozole for 23 days, BBR low (95mg/kg) and high (190mg/kg) doses administered for 4 weeks after PCOS induction. BBR ameliorated PCOS as evidenced by histological picture and modulating serum hormonal profile. Moreover, BBR hampered cAMP/protein kinase A (PKA) levels to attenuate abnormal steroidal enzymes (steroidogenic acute regulatory protein (StAR), CYP11A, CYP17A, CYP19A). In parallel, BBR induced an expressive decrement in the cAMP effector, the exchange protein directly activated by cAMP (Epac) and consequently phospholipase C (PLC), inositol triphosphate receptor (IP3R) as well as calpain levels to induce autophagy as evident by the marked rise in the microtubule-associated protein light chain 3 (LC3II/LC3I) ratio. Therefore, BBR could serve as a potential candidate for halting PCOS progression through interfering with cAMP action. It impedes both PKA and Epac axes which are master regulators of steroidogenesis and autophagy, an effect that could serve, in part, as a possible novel explanation for the beneficial role of BBR in treating PCOS.

Summary, etc. متلازمة تكيس المبايض المتعدد(PCOS) هو مرض منتشر ولا يوجد له علاج حتى اليوم. يتسبب احادي فوسفات الأدينوزين (cAMP) في زيادة تكوين الستيرويدات في المبيض و خفض الالتهام الذاتي.<br/>بربري (BBR) هو مشتق من نبات طبيعي تم استخدامه على مدار قرون طويلة كعلاج لتحريض الاباضة. تهدف هذه الدراسة الي تسليط الضوء الى دور البربرين في علاج متلازمة تكيس المبايض المتعدد عن طريق تقليل مستوى احادي فوسفات الادينوزين الحلقي.<br/>تم تقسيم 32 جرذاً من فصيلة ويستار البيضاء الى 4 مجموعات: مجموعة ظابطة, مجموعة مثلث نموذجاً للمتلازمة, مجموعة تلقت جرعة قليلة من البربرين(95 mg/kg) و مجموعة تلقت جرعة عالية من البربرين(190 mg/kg) .<br/>ادي علاج البربرين الي تحسين التشريح المبيضي المرضي و تقليل الانحرافات الهرمونية و ايضاً قلل البربرين التعبير عن الانزيمات المساعدة لتكوين الستيرويدات في المبيض (StAR, CYP11A and CYP17A) و نشاط الانزيم CYP19A <br/>اظهر العلاج بالبربرين انخفاضاً في مستوى بروتين التبادل المنشط بواسطة احادي فوسفات الادينوزين الحلقي (Epac) , فوسفوليباز (PLC), مستقبل الاينوزيتول ثلالثي الفوسفات (I3PR) و مؤشر الالتهام الذاتي كالباين و تعزيز نسبة سلسلة البروتين الخفيفة المرتبطة بالانيبيبات الدقيقة (LC3II/LC3I) و بالتالي يمكن استنتاج اهمية و دور البربرين في تقليل تقدم المتلازمة عن طريق التدخل في عمل احادي فوسفات الادينوزين الحلقي
