Background: Malathion is a widely used organophosphorus insecticide, however, its unregulated use have produced severe environmental pollution and potential health hazards. N-acetylcysteine (NAC) is a known glutathione precursor and nowadays there is a growing interest concerning its beneficial effects against pesticides-induced toxicity. Objectives: The present study was designed to investigate malathion-induced hepatotoxicity in the form of liver cell injury and histopathological changes in liver tissue, immunotoxicity and genotoxicity and also to clarify the possible causing mechanisms of these hazards and finally, to evaluate the protective effects of NAC against these hazards in rats that were exposed to low dose malathion for 90 consecutive days. Materials and Methods: 100 adult male albino rats were randomly divided into four groups comprised of 25 rats each. For 90 consecutive days, Group 1 received distilled water daily, Group 2 received NAC in a dose of 150 mg kg/day, Group 3 received malathion in a dose of 50 mg/kg/day and Group 4 received malathion in a dose of 50 mg/kg/day followed by NAC in a dose of 150 mg kg/day.
