Summary, etc. The present study aimed at investigating the therapeutic effect of SERCA2 gene transfected adipose mesenchymal stem cells (AMSCs) on induced wound in type1 diabetes mellitus (T1DM) in male albino rat model and comparing the effect to that of nontransfected AMSCs. Forty two adult male albino rats were divided into: Donor Group included 3 rats for stem cell isolation, culture, phenotyping, labeling and SERCA2 preparation. Group Ї included 9 rats. Group II (Diabetic Wounded Group) included 10 rats. Diabetes was induced by a single intraperitoneal (IP) injection of STZ at a dose of 50 mg/kg. The rats were exposed to wound induction (1x1cm skin incision) on the middle part of the back the day following confirmation of diabetes. Group III (Diabetic Wounded and Non-transfected AMSCs treated Group) included 10 rats. 1x106 of cultured and PKH26 labeled rat cells were injected IP. Group IV (Diabetic Wounded and SERCA2 transfected AMSCs treated Group) included 10 rats. 1x106 of cultured, green fluorescent protein (GFP) labeled and SERCA2 1.5 microgram (og) lipofectamine transfected were injected IP. The rats were sacrificed 3 weeks following the day of confirmation of diabetes. Blood glucose level, skin Ca2+ concentration, western blot analysis, quantitative polymerase chain reaction (qPCR), histological, immunohistochemical, fluorescent microscopic, morphometric and statistical studies were done The mean value of blood glucose indicated a significant increase in the diabetic group compared to the control and the treated groups. The mean Ca ion concentration and mean SERCA2 protein values indicated a significant decrease in the diabetic group compared to all other groups and in AMSCs group versus control and SERCA2 group. Group II demonstrated wide separation of the wound edges, detached parts of the epidermis, disrupted dermal fibers, detached sebaceous glands, follicles lined by 1 layer of cells, multiple microfollicles. Thickened and thinned wound edges, multiple vacuolated keratinocytes, decreased or loss of granular content of granular cell layer,disorganized areas and vacuoles were seen
