Summary: Altered mitochondrial energy metabolism is recently observed on cancer cell proliferation and numerous natural products exhibit cytotoxic effects against different cancer cells. Therefore, This study was done to Investigate the effect of Alchemilla vulgaris extract on Chromatin diffusion and mitochondrial ND3 gene expression in PaCa-2 human pancreatic cancer cell line. The effect of Alchemilla vulgaris extract on proliferation and viability of cancer PaCa-2 cells was studied using cytotoxicity MTT test. Chromatin diffusion assay was done to detect DNA damage induction, and also the expression level of mitochondrial ND3 gene was measured using RT PCR. Results of the MTT test revealed a concentration dependent death of PaCa-2 cells after treatment with different concentrations of Alchemilla vulgaris extract. High induction of DNA damage and remarkable over expression of mitochondrial ND3 gene were also induced after treatment of PaCa-2 cells with three different concentrations of Alchemilla vulgaris extract. Conclusion: Alchemilla vulgaris extract exhibited a strong concentration dependent cytotoxic effect against cancer PaCa-2 cells through high induction of DNA damage and marked overexpression of mitochondrial ND3 gene. Thus more studies are required to investigate of possibility of using Alchemilla vulgaris extract in cancer treatment.Summary: لوحظ تغيير استقلاب طاقة الميتوكندريا مؤخرا علي تكاثر الخلايا السرطانية والعديد من المنتجات الطبيعية تظهر تأثيرات سامة للخلايا ضد الخلايا السرطانية المختلفة . لذلك تم اجراء هذه الدراسة للتحقيق في تأثير مستخلص رجل الاسد علي انتشار الكروماتين والتعبير الجيني للميتوكندريا ( 3-ND) في خط خلايا سرطان البنكرياس البشري )2PaCa-) . تمت دراسة تأثير مستخلص رجل الاسد علي تكاثر الخلايا السرطانية (PaCa-2) وحيويتها باستخدام اختبار (MTT) للسمية الخلوية. تم اجراء اختبار انتشار الكروماتين للكشف عن تحريض تلف الحمض النووي وكذلك تم قياس مستوي التعبير عن جين ND-3 الميتوكندوري باستخدام ( (RT PCR . كشفت نتائج اختبار ال MTT عن موت معتمد علي التركيز لخلايا PaCa-2 بعد العلاج بتركيزات مختلفة من مستخلص رجل الاسد . كما تم تحريض عالي لتلف الحمض النووي والتعبير المفرط عن جين الميتوكندريا ND-3 بعد معالجة خلايا PaCa-2 بثلاث تركيزات مختلفة من مستخلص رجل الاسد . الخلاصة : اظهر مستخلص رجل الاسد تأثيرا ساما للخلايا يعتمد علي تركيز قوي ضد خلايا سرطان PaCa-2 من خلال الحث العالي لتلف الحمض النووي والتعبير المفرط لجين الميتودندريا ND-3 . وبالتالي هناك حاجة الي المزيد من الدراسات للتحقيق في امكانية استخدرام مستخلص رجل الاسد في علاج السرطان .
