This study was carried out at the Tissue Culture Laboratory , Vegetable Crops Department , Faculty of Agriculture , Cairo University , during the period from 2003 to 2005The main objective of this work was to study the effect of sodium azide (SA , NaN3) (0 , 05 , 075 and 1 mM) nd Gamma irradiation (0 , 10 , 20 , 30 , 40 Gy) on improving salinity tolerance of the Spunta and Diamant potato cultivars shoot tips in vitro , using sea salt concentrations (0 , 1000 , 2000 , 4000 and 8000 ppm) Also producing plants able to grow under high levels of salinity concentrations , aiming to induce and select in vitro salt tolerant plant linesAnother part of the study was to investigate the effect of gamma irradiation and SA on microtuberization of Spunta and Diamant potato cultivarsThe obtained results showed that low doses of gamma irradiation (10 and 20 Gy) improved growth parameters (plant length , number of leaves and survival percentage) in both of Spunta and Diamant cultivars at the three subculturesWhile the highest doses (30 and 40 Gy) adversely affected growth parametersAs for the effect of sea salt , 1000 ppm caused the highest value of plant length and number of leaves , while 8000 ppm showed the lowest value in the two cultivars Spunta and Diamant at the three subculturesThe interaction between gamma irradiation treatments 30 and 40 Gy with 8000 ppm sea salt , and the interaction between 075 mM SA concentration and 8000 ppm adversely affected growth parameters (plant length , number of leaves and survival percentage) , while the highest value of growth parameters were obtained from the interaction between irradiation treatments 10 and 20 Gy with 1000 ppm and 10 Gy with 2000 ppm
