The present investigation deals with the synthesis of 6-aryl-9-substituted-6,9-diazaspiro-[4,5]decane-8,10-diones (VIa-i) and 1-aryl-4-substituted -1,4-diazaspiro[5.5]undecane-3,5-diones (VIm-u), 6-aryl-9-(1H-tetrazol-5-yl)methyl)-6,9-diazaspiro[4.5]decane-8,10-diones (VIj-l) and 1-aryl-4-((1H-tetrazol-5-yl)methyl)-1,4-diazaspiro[5.5] undecane-3,5-diones (VIv-x). The anticonvulsant activity was studied using subcutaneous pentylenetetrazole (ScPTZ) test, Maximal electroshock seizure (MES) test and Neurotoxity test. The molecular modeling part was done through pharmacophore generation using Discovery Studio 2.5 software (Accelerys, Inc.: San Diego, CA.). The thesis is divided into the following sections: Introduction This section includes a review on the chemistry and biological profile of diketopiperazines (DKPs) in addition to the biological profiles of disubstituted cycloalkanes. Basis of the present investigation This part rationalizes and describes the pharmacological basis on which the synthesized compounds were selected to be studied as anticonvulsants, in addition to the schemes followed in their synthesis (Schemes 1-3)
