Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Veterinary Medicine Department of Microbiology Summary: In this study, 250 edible egg samples (shell, egg membranes, albumen and yolk ) collected from different sources were examined .The result of isolation of gram negative bacteria revealed that there were 89 isolates which biochemically identified as,40 Salmonella isolates (16%),18 Proteus isolates (7%),16 E coli isolates (6%) and15 Klebsiella isolates (6%). Serotyping for Salmonellae isolates revealed that there were 8 S. kentuky , 8 S. virchow ,7 S. Enteritidis ,7 S. Infantis ,5 S. Newport , 3 S. Typhimurium ,2 S. Hidelberg .The results of virulence study revealed that 15 Salmonella strains were positive for Congo red and death occurred to chick embryos inoculated by Salmonella after third day of inoculation as 100% in case of Salmonella Enteritidis , 80% in case of Salmonella Typhimurium. Most of Salmonella isolates revealed their resistance for the following antibiotics: (chloramphenicol, streptomycin, lincomycin). Confirmation of 40 recovered Salmonella serovars by PCR using 16srRNA specific primers. Molecular characterization for S. Enteritidis, S. Typhimurium on DNA extracted from eggs albumen samples after selective enrichment on RVs broth by using (sefa , flic) gene primers for each serovar respectively revealed that there were 12 positive samples for Salmonella Enteritidis and 5 positive samples for Salmonella Typhimurium. Studying of virulence genes as (yafd , xtha) on the 7 S. Enteritidis and 3 S. Typhimurium recovered from eggs revealed that both genes present in S. Enteritidis but only (xtha) gene present in S. Typhimurium. While studying the previous virulence genes on the DNA extracted from eggs albumen samples after selective enrichment on RVs broth revealed negative results
