Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Veterinary Medicine - Department of Virology Summary: This study presents a trail for preparation of AI-ND and AI vaccines and evaluate the immunogenicity and protective efficacy of such vaccines emulsified with Montanide ISA 71 adjuvant. Characterization of humoral and cellular immune response and challenge test were carried out in specific pathogen-free chickens. The virus shedding was detected in the swab samples collected post challenge by RT-PCR. The results revealed that a single immunization with the prepared vaccines induced early up regulation of IL6 gene expression, high levels of HI antibody titers, enhance the phagocytic activity and protected chickens against the challenge with the H9N2 and ND viruses. To our knowledge, this is first study in Egypt using Montanide ISA71 as adjuvant for inactivated LPAI H9N2 and ND (VIId strain) vaccine to enhance both cell mediated immune response side by side with acquired specific immune response
