Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Pharmacology and Toxicology Summary: Ischemia/reperfusion (I/R)-induced acute renal failure (ARF) causes renal dysfunction. The present study was designed to investigate the potential reno- protective and/or therapeutic effects of rebamipide against renal I/R in rats. Male Wistar rats were divided into normal, sham, I/R, prophylactic, and treated groups; the latter 2 groups received rebamipide (100mg/kg/day, p.o) for 14 days before or after the I/R insult. The ARF caused a significant increase in serum creatinine, BUN and cystatin C. It caused also a significant decrease in tissue total antioxidant capacity and GSH levels, with an elevation in the content/activity of ICAM-1, MPO and MDA. Similarly, contents of PGE2, KIM-1, caspase-3, NF-mB and IF-Þ, were elevated in the I/R group as compared to the control one. The two therapeutic regimens reversed the altered biomarkers and histopathological changes; the effects of the treatment regimen show better results than those mediated by the protective one. Findings of the present study prove that rebamipide exerts its reno-protective and therapeutic effects via inhibiting neutrophils infiltration and its anti- inflammatory, anti-apoptotic and antioxidant activities
