Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Medicine - Department of Clinical and Chemical Pathology Summary: Background:Acute myeloid leukaemia (AML) is considered a stem cell disease characterized by clonal growth and the accumulation of myelopoietic progenitor cells. Objectives:To investigate the expression pattern of CD96 and CD117 in bone marrow or peripheral blood from adult patients with AML and their value for diagnosis. Patients &Methods:The study was carried out on 40 patients with de novo AML(as patient group), as well as 40 bone marrow samples from patient with non-haematologicalmalignancies(as a control group).Using flow-cytometry, we analyzed the expression pattern ofCD96, CD117among CD34+/CD38- cells. Results:The Percentage of CD96+ cells among CD34+CD38- cellswas higher in AML cases ranged between 0.13%-35.2%(median 6.53%) than for control group which ranged between 0.01%-2.98%(median 0.49%)that show a highly statistically significant difference(p value < 0.001 in both).Also, Expression (mean fluorescent intensity) of CD96 was higher among CD34+CD38- cells in AML cases at diagnosis ranged between 0.12-18.5 (median 3.3) than for control group which ranged between 0.11-4.1 (median 1.78). The Percentage of CD117 cells was higher among CD34+ CD38- cells in AML cases at diagnosis rangedbetween 0.01%- 4.17%(median 0.58%)than for control group which ranged between 0.01%- 2.13% (median 0.36%),however this difference didn't show statistical significance (p =0.079)
