Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Biochemistry Summary: The present study was designed to investigate the ameliorative effects of curcumin, silybin phytosome® (ultrathistle®) and Ü-R-lipoic acid on thioacetamide (TAA)-induced chronic hepatitis and cirrhosis. For the induction of chronic hepatitis, rats were treated with TAA, administered intraperitoneally at a dose of 200 mg/Kg 3 times weekly for 4 weeks. At the end of this period, a group of rats was sacrificed to assess the development of chronic hepatitis in comparison to the respective control group. Chronic hepatitis rats were then allocated into two groups. In group (1), rats continued on TAA injection for additional 7 weeks. Meanwhile, in group (2), TAA administration was stopped and rats were subsequently subdivided into four subgroups. Subgroup (1) was left untreated, whereas subgroups from (2-4) were allowed to receive daily oral doses of either curcumin, ultrathistle® or Ü-R-lipoic acid, respectively for 7 weeks. On the other hand, rats for cirrhosis model were allocated into two groups of which group (1) received TAA for 7 weeks and group (2) was subdivided into three subgroups. Animals from subgroups (1-3) were given daily oral doses of curcumin, ultrathistle® or Ü-R-lipoic acid for 7 weeks simultaneously with TAA. Curcumin, ultrathistle® and Ü-R-lipoic acid displayed a variable degree of attenuation of TAA-induced chronic hepatitis and cirrhosis
