Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Pharmacology and Toxicology Summary: The present study was designed to investigate the effectiveness of rosiglitazone, gliclazide alone as well as in combination with fenugreek and vitamin E in treatment of type 2 diabetes mellitus either in STZ regimen. In addition, amelioration of the possible adverse effects of gliclazide and rosiglitazone by fenugreek and vitamin E were also assessed. Type 2 diabetes mellitus (T2DM) was induced in rats by a single intraperitoneal (i.p) injection of streptozotocin (45 mg/kg b.wt; STZ). Rosiglitazone (10mg/kg), gliclazide (10mg/kg), (fenugreek 100mg/kg) and vitamin E (200mg/kg) were administered daily for 14 days after induction of diabetes. At the end of experiment period ,blood and tissue samples were collected and blood glucose, glycosylated hemoglobin (HbA1c), insulin, lipid profile as (TC, TG, HDL-C, LDL-C), lipid peroxidases (MDA, GSH), liver function (ALT, AST), creatine kinase monoenzyme B(CK-MB), lactate dehydrogenase (LDH) as well as histopathological examination of heart were determined . In STZ regimen showed significantly elevated in all biochemical parameters except plasma insulin and GSH level was significantly decreased as compared to their respective control group and produced pathological alterations in examined tissues. Treatment of rats with fenugreek ,vitamin E and low doses of rosiglitazone and gliclazide significantly reduced blood glucose, HbA1c, TC, TG, HDL, LDL, MDA, ALT, AST, CK-MB and LDH levels and significantly elevated in GSH and plasma insulin levels as compared to diabetic group
