Aim: To evaluate permeability of dentin: 1. Treated with Tri-calcium phosphate varnish versus Fluoride-containing desensitizing varnish. 2. After a brushing-abrasion challenge of the treated dentin. Materials and Methods: A total of 124 freshly extracted sound human molars were sectioned into 0.5 mm thick dentin discs. The discs were randomly divided into two equal groups, 62 discs each, according to varnish used; tri-calcium phosphate varnish (Clinpro white varnish) and fluoride-containing desensitizing varnish (Topex Durachield). Each group was further subdivided into two subgroups, 31 specimens each, according to the exposure to mechanical challenge by tooth brushing; either subjected to mechanical abrasion by tooth brushing or not. Dentin permeability was measured at base line and after treatment and storage under simulated pulpal pressure for 10 minutes. Also scanning electron microscope (SEM) was performed. Results: On comparing the median percent reduction of dentin permeability after the application of the tested varnishes, tri-calcium phosphate varnish showed higher value than fluoride-containing varnish. However, the difference was not statistically significant (p = 0.942). It was also found that brushing-abrasion challenge did not significantly affect dentin permeability regardless the type of tested varnish (p = 0.732). On the other side when comparing the influence of brushing-abrasion challenge on the tested varnishes the difference was also not statistically significant (p = 0.617). These results were supported by scanning electron microscope (SEM). Conclusion: Irrespective of the varnishes used, occlusion of the dentinal tubules and reduction of dentin permeability did occur. Both types of varnishes are effective desensitizers and did reduce the dentin permeability after tooth brushing abrasion challenge.
