Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Science - Department of Zoology Summary: Cystic echinococcosis due to Echinococcus granulosus (E.granulosus) is one of the most important chronic helminthic diseases, especially in sheep/cattleraising regions. The larval stage of the parasite forms a cyst that grows in the liver, lung, or other organs of the host. Immunodiagnostic methods are more applicable for their better sensitivity and specificity. Cathepsin-B from sheep and camel livers was purified by ammonium sulfate fractionation and a series of chromatographies on DEAE Sepharose CL-6B and Sephadex G-75. The purified protein was injected in rabbits to raise specific polyclonal antibodies (pAb) against E.granulosus. Detection of cathepsin-B in serum of human by Sandwich-ELISA gave a sensitivity of 90.38%, a specificity of 91.95%. On the other hand, detection of cathepsin-B in serum of human by Dot-ELISA gave a sensitivity of 91.29%, a specificity of 92.34%. In conclusion, cathepsin-B antigen appear to be sufficiently sensitive and specific antigen for the detection of human and animal echinococcosis using sandwich- ELISA and Dot- ELISA
