This study have been conducted on 110 female albino rats (150-200g) divided into 11 groups (10 rats/group) for the purpose of testing the hepatoprotective effect of both inulin nanoparticles and silymarin against chlorpromazine-induced hepatotoxicity.It was detected throughout the study that various liver enzymes were seriously affected with chlorpromazine(CPZ) treatment while upon treatment with inulin nanoparticles whether in low or high doses and silymarin, a significant improvement was achieved. Significant increases in ALT, AST, ALP, LDH,LDL, TB, MDA, TNF, NO, cholesterol and triglycerides accompanied with a significant decrease in ATP, CAT, GPx, SOD and HDL were detected after (CPZ) treatment. Animals received inulin nanoparticles at the two tested doses and silymarin plus (CPZ) showed a significant improvement in all biochemical parameters toward the normal value of the controls. Moreover treatment with (CPZ) resulted in a significant decrease in glutathione peroxidase (GPX)and superoxide dismutase (SOD) level in liver accompanied with a significant increase in oxidative stress markers expressed as lipid peroxidation (MDA) and serum nitric oxide (NO) in the liver tissue. Treatment with inulin nanoparticles and silymarin succeeded to induce a significant improvement in antioxidant parameters and oxidative stress markers. The tumor marker tumor necrosis factor (TNF-Ü) was increased significantly in the group treated with (CPZ)alone while the combined treatment with the low or high doses of inulin nanoparticles or silymarin plus (CPZ)succeeded to induce a reduction in (TNF-Ü).The results also indicated that rats treated with (CPZ)showed a significant cytotoxicity as indicated by the depletion in DNA and RNA content in the hepatic tissue, increase DNA fragmentation which indicated the apoptotic effect of (CPZ)accompanied with a significant increase in tumor necrosis factor (TNF) gene expression in liver
