Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Veterinary Medicine - Department of Microbiology Summary: Chlamydia psittaciis a biological agent endemic in nearly all birds species and large and small ruminants, it has a severe zoonotic to effect to be transmitted to humans. Economic impact and financial losses resulting in from avian chlamydiosis especially those in pet birds, poultry and large and small ruminants combined with its zoonotic effect on humans and tranmitted to large numbers of people causing disease .It must and necessary of application of proper control measures. In the present study the potential risk of Chlamydia psittaci infection was determined by isolation of Chlamydia psittaciin excreta, nasal and pharyngeal and conjunctival samples of poultry species (Turkey, pigeons, chicken and duck). The prevalence of chlamydiosis using Gimenez stain was 74.45% in turkey,75.2% in pigeons, 17.5 in chickensand 5.62%in duck.The result of molecular identification was 30% so birds owner in farms could be under the risk of chlamydial infection. For the first time in Egypt, in the present study, preparation of inactivated vaccine against Chlamydia psittaci (local strain), was investigated. The embryonated chicken egg grown material of Chlamydia psittaci strains was used as a whole bacterin and propagation for purification in tissue culture (Vero cells) to vaccinate mice as a model. In Immunization experiment the protection after challenge was nearly 100%.The trial histopathological examination of internal organ and Giemsa stain of frozen organs revealed that the prepared killed vaccine can effectively reduce chlamydial infection in birds and animals with proper level of protection
