The present study aimed to determinetion the natural prevalence of infection with Anisakis simplex larvae (L3) infecting Dicentraches labrax in Egypt, followed by a detailed description of the recorded parasites by means of light and scanning electron micrioscopy. Nematode larvae were collected from the surface of visceral organs such as the stomach, intestine, and muscles; subsequently rinsed in phosphate-buffered saline (PBS); fixed in hot 70 % ethanol (60 {u00B0}C). Investigation and determination of cell mediated immune response (IFN-Þ, TNF-Ü, IL- 5, IL-4, IL-6 and IL-17) and nitric oxide (NO) concentration released against larvae (L3) antigens during an experimental infection into male Wistar rats by ELISA were carried out. Also, to assess the presence of total IgG and IgE responses against L3 antigens by ELISA. Forty male Wistar rats were divided into five groups (eight rats each):1.Control (non-infected and untreated) group 2. Fresh group rats were orally inoculated with freshly collected L3 larvae. 3. Frozen group rats were orally inoculated with L3 frozen at {u2212}20 {u00B0}C for 1 week. 4. Cooked group rats were orally inoculated with L3 cooked in 100 {u00B0}C water bath for 10 min. The larvae were cut into pieces before inoculation (three to five larvae per rat). 5. Injecte groups rats were sensitized by intraperitoneal injection of crude antigen extract emulsified in an complete Freund{u2019}s adjuvant (1:1 volume) containing 500 og/ml. The sensitized rats were then boosted with incomplete Freund{u2019}s adjuvant after 7 days
