Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Medicine - Department of Medical Biochemistry Summary: Background : Psoriasis is a common chronic, recurrent, immune mediated disease of the skin and joints. Sirt1 is a protein implicated in regulation of many cellular processes, including apoptosis, cellular senescence, endocrine signalling, glucose homeostasis, aging and longevity. Autophagy is the process by which dysfunctional cellular components are degraded inside the cell through the action of lysosomes. Aim of this work: The aim of this work is to detect the expression level of sirt1 in psoriasis cases and to study its association with autophagy. Patients and Methods: The expression of sirt1 and beclin1 were detected in 30 psoriasis patients and 25 healthy controls using quantitative real-time PCR. Results: The expression of sirt-1 was significantly lower in cases rather than in the controls. Also, the expression of beclin1 was significantly lower in cases group rather than in the control group. Correlation was positive between expression level of sirt1 and beclin1 in psoriasis patients, and negative in healthy group. Conclusion: Sirt1 may play a role in pathogenesis of psoriasis, which may be through autophagy
