Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Medicine - Department of Clinical and Chemical Pathology Summary: Background:In Egypt, CRC is the 5th most common malignant tumor representing 5% of the total estimated cancers according to national cancer registry 2013. The aim of the study was to evaluate associations of serum level of Cyclin D1 and variant in MIR196A2 rs11614913 and DCC rs714 genes with the risk of colorectal cancer. It aimed to correlate genotypes with tumor site, stage, histopathological grade as well as lymph node involvement and distant metastasis. Subject and methods:The serum level of Cyclin D1 was measured by ELISA. The DCC SNP was assayed by PCR-RFLP method and MIR196A2 SNP by Real-Time Taqman assay, in 100 colorectal cancer patients selected from the Oncology department, Kasr Al Eini, Cairo University and 60 healthy age-matched controls. Results:A significant association between increase the serum level of Cyclin D1 and risk of colorectal cancer (pvalue=0.001) with cut-off >3.4. Mutant variant genotypesAG, GG of DCC were associated with increase the risk of colorectal cancer compared with AA genotype (pvalue<0.001, OR 3.1, 95% CI= 0.947 {u2013} 10.149).There was a significant increase in the risk of colorectal cancer in patients carrying the G allele compared to A allele (pvalue<0.001,OR= 2.591, 95% CI =1.59-4.206) also, DCC GG genotype were associated with increased the risk of distant metastasis in patient of CRC compared to DCC AA genotype (pvalue=0.014).Mutant variant CC, CT of MIR196A2 were not significantly associated with increase the risk of CRC (pvalue=0.766, OR=1.368, 95% CI=0.498- 5.074) but showed a significant increase in the progression of the tumor (pvalue=0.013) compared to TT genotype. Conclusion: Mutant variant GG and AG of DCC and increase serum level of Cyclin D1 are predictive factors of sporadic colorectal cancer in Egyptian patients. CC and CT genotype of MIR196A2 and GG genotype of DCC are markers of poor prognosis in patients of CRC
