Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Pharmaceutics Summary: In this work, for the first time, meropenem-loaded chitosan nanoparticles were prepared and evaluated as a potential tool to overcome antibacterial resistance, to improve pharmacokinetics of the drug and to control its release, and thus reducing the frequency of administration, with a subsequent increase in patient compliance. Thus, the work in this thesis was divided into:Chapter I: Preparation and Characterization of Meropenem-loaded Chitosan Nanoparticles. Chapter II:In vitro and In vivo Evaluation of Antibacterial activity of Meropenem-loaded Chitosan Nanoparticles. And these are the research findings:{u2022}Spherical nanosized meropenem-loaded CS nanoparticles with positive Zeta-potential values were prepared and demonstrated ultrahigh encapsulation efficiency of meropenem.{u2022}The release of meropenem from CS nanoparticles was characterized by a biphasic release pattern and controlled by a Fickian diffusion mechanism.{u2022}Meropenem-loaded nanoparticles with CS: TPP weight ratio of 5:1 showed optimum EE and controlled release profile.Thus, it was selected for further in vitro and in vivo antibacterial activity studies. {u2022}Higher antibacterial activity of meropenem-loaded CS nanoparticles was observed in vitro against clinical isolates of multidrug-resistant E. coli, K. pneumoniae and MRSA, as compared to free drug
