Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Medicine - Department of Histology Summary: Background:Acute pancreatitis (AP)is one of the most common digestive system inflammatory disorders leading to hospital admission with increasing annual incidence.Mesenchymal stem cells (MSCs) & Wheat germ oil (WGO) could have a therapeutic role through theiranti-inflammatory & antioxidant properties. Objective:To evaluate and compare the possible therapeutic effects of Bone marrow derived MesenchymalStem Cells (BMSCs) versus WGO on AP. Methods:Forty-seven adult male albino rats were included in the experiment. Twelveratsserved as control group I (no.=12). AP was inducedin the remaining thirty five rats by a single intraperitoneal (I.P.) injection of L-arginine (250 mg/100g), five rats died within the 1st hour after AP induction, the rest were divided randomly in to groupII (acutepancreatitis group; no.=10) receive no treatment , groupIII (stem cell treated group(BMSCs);no.=10) and groupIV(Wheat germ oil group (WGO);no.=10).One hour after AP induction group III was injectedI.P. by 1ml of PKH26 labeled BMSCs suspension in Phosphate Buffer Saline (PBS) (1x106 cells/ml),group IVreceived WGO in a dose of 3 ml/kg body weight by oral gavage every 24 h for 3 successive days.Bloodsamples were collected24 hours and on the 4thday (at the endof the experiment) after AP induction for biochemical assessment of serum amylase, lipase, interleukin-1Ý and interleukin-10. At the end of theexperiment, animals were sacrificed and specimens from the pancreas were taken and prepared forHematoxylin& eosin (H&E) stainandimmune-histochemical staining usinginducible Nitric Oxide Synthase (iNOS) and insulin antibodies.Morphometric measurements using image analyzer were done
