In addition, smoking is associated with potentially malignant disorders of the oral and oropharyngeal mucosa, however, when it comes to OLP, little is known about the influence of smoking on its potential of malignant transformation. Accordingly, in our study we aimed to evaluate the effect of smoking on ABCG2 and TP53 expression in OLP as markers for malignant transformation.Four groups of subjects were included in our study (10 in each group), nonsmoker/ control, smoker/control, non-smoker/OLP and smoker/OLP groups.The mucosal biopsies were stained by routine H&E staining and examined under the ordinary light microscope.The biopsies were immuno-stained by ABCG2 and TP53. The area percent for ABCG2 immuno-expression and the cell count for TP53 immuno-expression were measured by the image analyzer computer system. Data was collected and expressed as means and standard deviation.The present work demonstrated that a statistically significant difference in TP53 expression was present between all the studied groups with the highest expression in the smoker/OLP group. However, no statistically significant variation was present in ABCG2 expression. Hence, it was concluded that there is a malignant potential in OLP which is exaggerated by the effect of smoking and ABCG2 is not a reliable marker
