Primary open-angle glaucoma is the most common type of Glaucoma. This work tries to determine the efficacy of L-Carnitine and Omega-3 fatty acids against oxidative damage and modulation in glaucoma. Ocular hypertension will be induced by injecting Betamethasone into the anterior chamber of the eye.Thirty rabbits were used and divided into five groups such as Group 1: Control, Group 2: Glaucoma, Group 3: Glaucoma treated with L-Carnitine. Group 4: Glaucoma treated with Omega-3 fatty acids. Group 5: Glaucoma treated with both L-Carnitine and Omega-3 fatty acids. Intraocular pressure values were determined by tonometry. The activities of Catalase, superoxide dismutase and reduced glutathione were assessed. The levels of malondialdehyde and apoptotic marker Caspase-3 were also measured.The obtained results showed a significant decrease in intraocular pressure in glaucoma rabbits treated with L-Carnitine and Omega-3 fatty acids. A significant increase in the levels of reduced glutathione, catalase and super oxide dismutase activities was observed in the rabbits treated with L-Carnitine and Omega-3 fatty acids compared to glaucoma group. The levels of malondialdehyde were decreased after L-Carnitine and Omega-3 fatty acids administration, moreover; apoptotic marker caspase-3 levels were decreased after L-Carnitine and Omega-3 fatty acids administration
