Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Medicine - Department of Parasitology Summary: The present work was conducted on 380 laboratory bred Swiss albino male mice to assess the effect of G.glabra and M.fragrans on acute and chronic toxoplasmosis in comparison to spiramycin. The acute infection was done by IP injection of T.gondii tachyzoites. The mice were treated 5 days then sacrified. The effect of drugs was assessed by the number of tachyzoites in the IP fluid, histopathological examination of liver tissue sections and SEM of IP fluid. There was a significant decrease in the number of tachyzoites detected in the IP fluid between the control group and all treated groups (P<0.05) and between the M.fragrans treated groups and spiramycin treated group, marked improvement of inflammation in all treated groups in addition to severe deformities and distortion in the T.gondii tachyzoites in treated groups observed by the SEM. The chronic group was infected by oral inoculation of T.gondii tissue cysts, treated for 14 days starting from the 4{u036D} {u036A} week p.i. and sacrified at the 10{u036D}{u036D} {u036A} week p.i. The effect of drugs was assessed by the number of tissue cysts in homogenized brain tissues, histopathological examination and SEM of brain tissue. There was a significant decrease in the number of tissue cysts detected in the brain between the control group and all treated groups (P<0.05) and between the M.fragrans treated groups and spiramycin treated group. There was also a significant difference between the group treated with G.glabra at a dose of 500mg/kg body weight loaded on CSNP in combination to spiramycin and the spiramycin treated group. Histopathological examination revealed marked improvement of inflammation in all treated groups and the SEM of brain tissues revealed remarkable changes in the shape and structure of the T.gondii tissue cysts in treated groups
