Background and objectives:Testicular torsion is an emergency condition that requires urgent surgical detorsion to avoid testicular necrosis. This study was designed to evaluate and compare the possible effects of metformin versus selenium on testicularischemia/reperfusion(I/R) injury in a rat model. Materials and Methods: Forty-eight adult male albino rats were used in this study, 16 rats served as sham-operated control (I), the remaining 32 rats divided equally into 4 groups: (II) ischemic, (III)I/R, (IV) I/R+ Metformin and (V) I/R+ Selenium. GIIwas subjected to 1-hour of ischemia then sacrificed. GIIIwas subjected to 1-hour of ischemia and 7 days of reperfusion then sacrificed. GIV and GVwere subjected to I/R injury, GIV received 100 mg/kg/day of metformin by oral gavage and GV received an intraperitoneal injection of 0.5 mg/kg/day of selenium, for 7 consecutive days then sacrificed. Just before sacrifice, blood samples were drawn and serum levels of malondialdehyde(MDA), glutathione(GSH)and testosterone were measured. Left testicular sections were stained with H&E, PAS and immunohistochemical stains for caspase-3 and connexin43 (CX43).Morphometric and statistical studieswere done
