Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Science - Department of Zoology Summary: Pancreatic polypeptide (PP) is a peptide hormone associated with the Nonfunctional Pancreatic Neuroendocrine Tumor (NF-pNET).Therefore, the quantification of the PP is of high importance for early detection of the NFpNET.Herein, aptamer probes for specific recognition of the PP were generated using magnetic beads based-SELEX. The Sanger sequencing technology was used to monitor SELEX, and bioinformatics tools were used to identify the enriched aptamers.A group of aptamers were generated that can bind to PP specifically with dissociation constants (Kd) in the nanomolar range. Moreover, one of the most promising aptamers (S12) was integrated with gold nanoparticles for building a colorimetric sensing platform for quantifying PP. The developed biosensor showed a wide linear dynamic range with 521 pM detection limit. Furthermore, the performance of the aptasensor was validated for the detection of PP in human serum and showed recovery values ranging between 92.4 and 93.9%. Therefore, this aptasensor has a great potential as a diagnostic tool for NF-pNETs
