Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Analytical Chemistry
This work is concerned with the quantitative determination of some dugs containing heterocycle group such as Ramipril in combination with either Amlodipine besylate or Atorvastatin calcium together with Aspirin in their co-formulated binary and ternary mixtures; respectively, and Benproperine phosphate as a single drug. Stability study was performed on Ramipril, Amlodipine and Atorvastatin under different stress conditions, then the obtained degradation products were isolated, purified, identified and were further involved in the applied techniques. Different analytical techniques were adopted, namely separation techniques including; chromatographic separations with HPLC and TLC-densitomety, in addition to capillary zone electrophoresis. Also spectroscopic methods mainly spectrofluorimetry and multivariate spectroscopic ones. In addition to electrochemical methods. Those methods were applied for the determination of the previously mentioned compounds in their pure form, laboratory prepared mixtures and dosage forms. Different experimental conditions were studied till reaching the optimum conditions. In part I: General introduction about heterocycles and different pharmacological actions of the selected drugs were summarized, in addition to the stability studies on the selected drugs. In part II: Different separation techniques were conducted including stability indicating HPLC and TLC on the binary mixture of RAM and AML in the presence of their obtained degradation products. For the ternary mixture of RAM, ATV and ASP stability indicating HPLC and TLC in addition to capillary zone electrophoresis were performed in the presence of their degradation products. In part III: Different spectroscopic methods were performed including spectrofluorimetric method on the binary mixture of RAM and AML. In addition to multivariate spectroscopic methods on both mixtures in the presence of their degradation products. In part IV: An Electrochemical method namely ; ISEs was applied for studying the potentiometric behavior of BN and its application to spiked human plasma. Also the fabricated sensor was used for on-line in-vitro potentiometric monitoring of BN release from its resinate formulated suspension.
