Thesis (Ph.D.) - Cairo University - Faculty of Science - Department of Botany and Microbiology
The present study was conducted to explore the biodiversity and study the activity of plant growth promoting (PGP) spore forming bacilli (SFB) associated with maize grown in 14 governorates in Egypt. A total of 147 SFB were isolated and primary screened for different PGP characteristics (indole acetic acid, phosphate solubilization, NH3 and HCN production). The phylogenetic diversity among bacterial isolates was analyzed by amplified ribosomal DNA restriction analysis. Ten ribogroups were identified using ARDRA.Based on PCR-ribotyping and in vitro PGP attributes, 45 potential isolates were selected for 16S rRNA sequence analysis. The 16S rRNA-sequence analysis of bacterial representatives from different ribogroups revealed that, majority of isolated bacteria (80%) was assigned to the Bacillus genus. These results strongly support that, SFB characterized in this study could be used as potential bacterial inoculants to promote plant growth and control plant pathogenic fungi
