Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Pharmacology and Toxicology Summary: The present study aimed to evaluate the effects of combining on 17Ý-estradiol (E2) with agomelatine (Ago) on depressive- and anxiety-like behaviors in young adult female rats induced by ovariectomy (OVX).Female rats were subjected to either sham operation (SO) or OVX; allowed to rest for one-week post-surgery, then divided into five groups; SO and OVX rats were administered saline orally (p.o.) and corn oil subcutaneously (s.c.) and served as the control group. OVX rats were treated with either Ago alone (40 mg/kg/day, p.o.) for 10 days, or E2 alone (40 æg/kg/day, s.c.) on the 13th and 16th day (24 h prior to anxiety and depression testing) or combination of Ago and E2. Elevated plus maze test (EPM) and forced swimming test (FST) were carried out on the 14th and 17th day, two weeks after surgery to ensure the induction of anxiety and depression with OVX, respectively. These tests were preceded by an open field test (OFT) to exclude nonspecific effects on locomotor activity in FST.Twenty-four hours after the behavioral test, the animals were anesthetized with thiopental (5 mg/kg, i.p.) and blood samples were collected to estimate estrogen levels
