Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Oral and Dental Medicine - Department of Dental Materials Summary: Aim: The aim of the present study was to evaluate and compare the cytotoxicity and antibacterial effect against Enterococcus faecalis of bleached turmeric hydro-alcoholic extract (intervention) versus sodium hypochlorite (control) endodontic irrigants, in addition to their effects on dentin microhardness. Methodology: Cytotoxicity was evaluated on human skin fibroblasts using MTT assay. The irrigants were tested at 1, 5 and 15 minutes time intervals. After contact time, MTT solution was added and well plates were incubated. After the incubation period, optical density was read and correlated with cell viability percent.Antibacterial efficiency was evaluated using direct contact test. Each endodontic irrigant was added to fresh E.faecalis suspension and brain heart infusion media then incubated for 48 hours. After incubation period, optical density readings were obtained and by ELISA reader at 620 nm.For the evaluation of microhardness, a total of 14 teeth were used. Mechanical preparation with intervening irrigation using the tested irrigating solutions was done. Each tooth was then sectioned vertically into two halves and equally divided into two groups to be immersed in the corresponding irrigant solution. VHN was recorded before and after immersion. Statistical analysis of data obtained from each test was performed (p<0.05). Results: Results of cytotoxicity test revealed that the intervention has significant more cell viability percent than the control at all time intervals (p<0.001). However, in the intervention group, cell viability percent significantly decreased over time.Results of antibacterial test showed bacterial inhibition by both groups. However, control group showed significantly higher bacterial inhibition (p>0.05).Results of microhardness test revealed that both groups showed significant reduction in microhardness of root dentin after treatment. Moreover, there was no significant difference between the mean differences of both groups before and after treatment indicating similar effect on dentin hardness
