Aim: The current research was carried out in order to study and compare the modulatory effect of H1 receptor antagonist first and second generations administration on alveolar bone healing following tooth extraction where H1 receptor antagonist drugs are commonly used worldwide and are affordable, so ensuring their bone regenerative effect following tooth extraction will be of great benefit to both patients and practitioners as well. Methodology : In the present study 24 adult male albino rats had their lower first molar extracted, 8 rats as a Control group, 8 rats as a Histaloc group and 8 rats as a Zyrtec group Alveolar bone healing after 3 and 7 days following tooth extraction was assessed by histological and histomorphometric analysis as well as PCR for Runx2 and FGF2.Results: Alveolar bone healing after 3 and 7 days following tooth extraction showed retarded healing in both first and second generations H1 receptor antagonist compared to the control group. In addition to, lesser amount of bone formation and lesser FGF2 and Runx2 expresion levels were detected.Conclusions: In the present experiment and according to the results, H1 receptor antagonist of both first and second generations negatively affected alveolar bone healing following extraction thus ; it's preferred to avoid its administration during the early stages of alveolar bone healing
