Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of African Postgraduate Studies - Department of Natural Resources Summary: Bovine tuberculosis is a zoonotic disease that not only causes huge economic losses but also poses an important risk for human infection. Consumption of unpasteurized milk can lead to the development of extra pulmonary TB.In the present study, a number of 2913 animals from milk farms in Cairo, El-Sharkia and El- Qalubia Governorates were examined via using of single intradermal cervical tuberculin technique (SICTT) and the results revealed that out of 2913 dairy cows, 108 dairy cows gave positive reactors with percentage (3.7%). The highest results for tuberculin skin test recorded in El - Sharkia Governorate (5.1%), then in El - Qalubia Governorate (4.2 %). While the result for tuberculin skin test in Cairo Governorate was (2.5 %).Thirty workers on the affected dairy farms were tested using a Mantoux test and the results revealed that none of 30 tuberculin reactive workers developed active tuberculosis.Two thousand eight hundred and five milk samples were collected from dairy cows farms from tuberculin tested animals .One hundred and eight lymph nodes samples were collected from positive cases. Samples were examined by culturing the prepared samples on Lowenstein Jensen media and middle brook 7H10 agar media.The results revealed 52 isolates of Mycobacterium spp. From milk samples were recovered on Lowenstein- Jensen media and middle brook 7H10 agar media with a percentage of isolation reached (1.85%) and 56 isolates of Mycobacterium spp. From lymph nodes samples were recovered on Lowenstein- Jensen media and middle brook 7H10 agar media with a percentage of isolation reached (51.85%). Also, all cultivated samples were confirmed by microscopic examination (using the modified ZiehlNelseen stain) and biochemical examinations
