Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Industrial Pharmacy Summary: The process of wound healing and tissue regeneration involves several key mechanisms to ensure the production of new tissues with similar cellular functions. The skin is the biggest organ and first barrier for the human being. Following a tissue injury, a systemic process occurs compromising four key elements; which are hemostasis, inflammation, proliferation, and maturation.These steps{u2019} regulation becomes affected with the variety of physiological and pathological conditions like age, infection or diabetes. Thus, the presence of a tissue regeneration promoter would be effective in accelerating wound healing process. Nitric oxide (NO) is a small diffusible molecule essential in regulating cellular and molecular functions. It is endogenously synthesized by NO synthase enzyme (NOS), and was found at elevated levels in wounds in healthy patients, and was deficient in some pathological conditions, such as non-healing chronic wounds, ischemic diseases, and carcinogenesis.This indicates its importance in the tissue healing process.Thus, the presence of a NO donor would be useful in accelerating the healing process. Nebivolol hydrochloride (NVH) is a third-generation beta-blocker approved by the food and drug administration (FDA) for the treatment of hypertension. NVH has been recently used for its tissue regenerating abilities, due to its NO donor ability. It exerts its vasodilating effects via NO pathway by releasing cardiovascular endothelial NO, in addition to its conventional beta-blocking effects. NVH causes endothelium dependent vasodilatation associated with the activation of L-arginine/NO pathway in both hypertensive and normotensive patients
