Owing to the poor outcomes and adverse side effects of existing ulcerative colitis drugs, developing an alternative nano-based treatment approach is highly warranted. Therefore, this proposal was designed to characterize the in vitro and in vivo properties of taurine, taurine-loaded chitosan pectin nanoparticles (Tau-CS-PT-NPs), and chitosan pectin nanoparticles (CS-PT-NPs) in the treatment of colitis induced by acetic acid in rats. For this purpose, CS-PT-NPs and Tau-CS-PT-NPs were prepared using the ionic gelation method and characterized by transmission electron microscopy (TEM), polydispersity index (PDI), zeta potential, Fourier transform infrared (FTIR) spectroscopy, encapsulation efficiency (EE), and drug release profile. Following colitis induction, rats were treated orally with free taurine, Tau-CS-PT-NPs, and CS-PT-NPs once per day for six days. Data showed that the size of Tau-CS-PT-NPs significantly increased compared to CS-PT-NPs. PDI and zeta potential values were 0.419, +48.4mV for CS-PT-Tau-NPs and 0.523, +43.5mV for CS-PT-NPs. EE was about 69.09±1.58%, and 60% of taurine was released in 4 h in simulated colon content. Acetic acid-induced colitis in untreated rats caused necrosis of colon tissues and a significant increase in interleukin-1beta, tumor necrosis factor-alpha, myeloperoxidase, and malondialdehyde levels associated with a remarkable decrease in reduced glutathione level in colon tissue in comparison to the control group, while, taurine, Tau-CS-PT-NPs, and CS-PT-NPs treatment partly reversed these effects. Our present study demonstrated that the administration of free taurine, CS-PT-NPs, and Tau-CS-PT-NPs exerted beneficial effects in acetic acid-induced colitis by their anti-inflammatory and antioxidant activities.Overall, taurine-loaded chitosan pectin nanoparticles emerge as a promising therapeutic option for colitis
