Thesis (Ph.D.) - Cairo University - Faculty of Pharmacy - Department of Pharmaceutics
Dapoxetine is a selective serotonin reuptake inhibitor. It is used for the treatment of the depression and premature ejaculation. It should be taken 2{u2013} 3 hours before sexual activity. Its maximum plasma concentration (Cm) is reached 2 hours after oral administration. Dapoxetine is metabolized extensively in the liver and kidney by multiple enzymes such as CyP2D6, CyP3A4, and flavin monooxygenase 1 (FMO1).The major product at the end of the metabolic pathway is circulating dapoxetine Noxide, which is a weak SSRI and contributes no clinical effect. Also, one of the most frequently encountered problems in the oral dapoxetine formulations is the bitter taste. Accordingly, the aim of the research is to prepare various taste-masked fast disintegrating orodispersible formulations in order to offer a faster dissolution rate, rapid onset of action, lower liver metabolism and better patient compliance
