Background: Burn is a major cause of traumatic injury to the human body causing tissue damage. Heat, chemicals, electric current, and other factors can all contribute to it. It primarily affects the skin and mucous membranes, but it can also affect muscles, bones, and even organs in severe cases. After a burn injury, infection and organ damage are the most common causes of death. Aim of the work: The aim of this study was to assess and compare the single and combined effects of SVF and PRP on the healing of a second-degree skin burn induced in male albino rats. Materials and Methods: This study included 46 male albino rats aged 8 to 12 weeks and weighing 150 to 175 grams on average. They were divided into the following groups: Control group (6 rats): healthy rats without 2nd degree skin burn wound induction. Group 2 (16 rats): rats with 2nd degree skin burn wounds were subdivided into 2 subgroups: 2a which was sacrificed at day zero to confirm the induction of 2nd degree skin burn and 2b where wounds were treated with local saline injections 500 oL at wounds margins and then sacrificed after one week.Group 3 (24 rats): rats with treated 2nd degree skin burn wounds were subdivided into 3 subgroups: 3a: rats were injected intradermally by 500 oL of SVF in saline solution (40 million cells per rat), 3b: rats were injected intradermally by 500 oL of PRP in saline solution, 3c: rats were injected intradermally by 1 mL of SVF&PRP then rats were sacrificed after one week.Results: The treated groups showed significant improvement in epidermis regeneration, increased collagen deposition and increased E-cadherin immune reactivity as compared to non -treated groups. The combined treated groups showed the best results when measuring the mean epidermal thickness, the mean area % of collagen deposition and E-cadherin as compared to SVF treated and PRP treated groups with no significant difference to the control group. In addition, there was no significant difference between SVF treated group and PRP treated group.
