The aim of the present in vivo study was the detection, quantitation and viability state of E. faecalis present in infected root canals before and after chemomechanical preparation using MTAD as an irrigant by culture and sensitive molecular techniques. A total of 50 patients of both sexes of age ranging from 18 to 40 years were enrolled. The patients required endodontic treatment for single rooted teeth. Patients were medically free and the teeth were necrotic asymptomatic with periapical lesions. Access cavities were made and samples were taken from the canals with paper points and transferred to the transport medium (S1 samples). S2 samples were taken from the canals after mechanical preparation and irrigation with 1.3% sodium hypochlorite and MTAD as a final rinse and then transferred to the transport medium. In the first part of this study, samples (S1 and S2) were taken to the microbiology lab for culture on CLED agar for detection and viability state of the bacteria. Subculturing was then done on EBA agar which is a selective media for E. faecalis. Gram stained film and catalase test were done for identification of the isolated bacteria
